    Large mammal remains from the early pleistocene site of Podere San Lorenzo (Perugia, Central Italy)

    Most of the research on fossil mammals from Umbria (central Italy) has been carried out in the southwestern branch of the Tiber basin, due to its paleontological richness. This portion of the basin extends from Perugia to Terni and corresponds to a well-defined half-graben filled with fluvial-lacustrine deposits. The paleontological sample presented here was discovered in a sand and gravel quarry at Podere San Lorenzo, East of the town of Deruta. The stratigraphic succession exposed in the quarry is no longer visible, but we describe here a new outcrop (Palazzone), which is not far from Podere San Lorenzo and shows comparable facies associations. The two successions were deposited in a fluvial environment characterized by an average reduction of the hydrodynamic energy from the bottom upwards. They are referred to the Early Pleistocene Santa Maria di Ciciliano Subsyntheme (Madonna dei Bagni Lithofacies). Large mammal remains are attributed to Mammuthus cf. meridionalis (Nesti, 1825), Stephanorhinus etruscus (Falconer, 1859), Equus stenonis Cocchi, 1867, Leptobos cf. etruscus (Falconer, 1868), ‘Pseudodama’ nestii (Azzaroli, 1947), and Sus strozzii Forsyth Major, 1881. Some hyena coprolites are also reported. The assemblage is typical of the early Late Villafranchian Land Mammal Age and can be referred to the Olivola/Tasso Faunal Units (about 2.0–1.8 Ma). This is in agreement with the alleged age of some other assemblages found in the southwestern branch of the Tiber basin (e.g., Torre Picchio, Villa San Faustino, Colle Sant’Andrea, Pantalla)

    Sensitization and Clinically Relevant Allergy to Hair Dyes and Clothes from Black Henna Tattoos: Do People Know the Risk? An Uncommon Serious Case and a Review of the Literature

    Henna (Lawsonia inermis L.) tattooing has been used in Egypt and India since ancient times. Today this temporary body art is becoming increasingly popular among young people. Various chemicals are added to henna to darken and enhance the definition of tattoos, especially para-phenylenediamine (PPD), which is a strong sensitizer known to cause cross sensitive reactions to azoic dyes and other para-amino compounds. We present the case of an 18-year-old girl who became clinically sensitive to textile dyes after having showed a serious reaction both to her first hair dying when she was 16 years old and following the application of a temporary henna tattoo when she was a kid. The evidence from our literature review showed 33 cases of manifest sensitization to hair dye and only one of observable contact allergy to both hair and textile dyes from henna tattoos. The sensitization of children may have long-life lasting consequences, because of cross-reaction to dyes and other chemicals contained in hair colourants, clothes and drugs. Since tattoos are very popular and globalization has increased the circulation of unauthorized products we point out the need for informative campaigns about the risk of sensitization caused by temporary tattoos

    Impact of aerosol microphysical properties on mass scattering cross sections

    We assessed the sensitivity of simulated mass scattering cross sections (αλsca [m2/g]) of three aerosol perturbed particle microphysical properties and derived constraintspecies to on these microphysical properties, suitable for the north-western Mediterranean basin, from a comparison between code calculations and observations. In detail, we calculated αλsca of mineral dust,organic carbon and sulfate at three wavelengths in the visible range with a T-matrix optical code, considering ±20%perturbations on size distribution, refractive index and mass density, and spheroids with two different axial ratios as shape perturbations.     From air quality to climate: Impact of aerosol sources on optical properties at urban, regional and continental levels in the north-western Mediterranean

    Further research is needed to reduce the existing uncertainties on the effect that specific aerosol sources have on radiative forcing, thus supporting the assessment of future mitigation strategies which should be focused on both air quality and climate, and not acting separately. This study presents a new approach aimed at quantifying the mass scattering and absorption efficiencies (MSE and MAE) of different aerosol sources at urban (Barcelona-BCN), regional (Montseny-MSY) and remote (Montsec-MSA) background sites in the northwestern (NW) Mediterranean. An analysis of source apportionment to the measured scattering and absorption coefficients was performed by means of a multilinear regression (MLR) model during 2010–2014 at BCN and MSY and during 2011–2014 at MSA. The source contributions to PM10 mass, identified by means of the Positive Matrix Factorization (PMF) model, were used as dependent variables in the MLR model in order to take into account the internal mixing state of atmospheric aerosols. Seven aerosol sources were obtained at MSA and MSY and 8 sources at BCN. Mineral, Aged marine, Ammonium sulfate, Ammonium nitrate and V-Ni bearing sources were common at the three sites. Traffic, Industrial/metallurgy and Road-resuspension were isolated at BCN, whereas Industrial/Traffic and Aged organics were solely identified at MSY and MSA. The highest MSE were found for Ammonium sulfate (4.5 and 10.7 m2 g−1), Ammonium nitrate (8.8 and 7.8 m2 g−1) and V-Ni (8 and 3.5 m2 g−1) at MSY and MSA respectively, dominating the scattering throughout the year with marked seasonal trends. V-Ni bearing, originated mainly from shipping emissions, simultaneously contributed to both scattering and absorption being the second most efficient absorptive source in BCN (0.9 m2 g−1). The Traffic source at BCN and the equivalent Industrial/Traffic at MSY and MSA mainly governed the light absorption and exhibited the highest MAE (1.7, 0.9 and 0.2 m2 g−1, respectively). Sources predominantly composed by fine and relatively dark particles such as Industrial/Traffic, Aged organics and V-Ni were simultaneously characterized with low single scattering albedo (SSA) and high scattering Angstrom exponent (SAE). Conversely, Mineral and Aged marine showed the highest SAE and the lowest SSA, being scattering the dominant process in the light extinction. The good agreement between modeled and measured optical properties allowed for the reconstruction of scattering, absorption and SSA time series by means of the PMF-MLR technique for the period 2004–2014 at MSY. Significant decreasing trends were found for the modeled scattering and absorption (−4.6 and −4.1 % y−1) coefficients.     Plzf Regulates Germline Progenitor Self-Renewal by Opposing mTORC1

    SummaryHyperactivity of mTORC1, a key mediator of cell growth, leads to stem cell depletion, although the underlying mechanisms are poorly defined. Using spermatogonial progenitor cells (SPCs) as a model system, we show that mTORC1 impairs stem cell maintenance by a negative feedback from mTORC1 to receptors required to transduce niche-derived signals. We find that SPCs lacking Plzf, a transcription factor essential for SPC maintenance, have enhanced mTORC1 activity. Aberrant mTORC1 activation in Plzf −/− SPCs inhibits their response to GDNF, a growth factor critical for SPC self-renewal, via negative feedback at the level of the GDNF receptor. Plzf opposes mTORC1 activity by inducing expression of the mTORC1 inhibitor Redd1. Thus, we identify the mTORC1-Plzf functional interaction as a critical rheostat for maintenance of the spermatogonial pool and propose a model whereby negative feedback from mTORC1 to the GDNF receptor balances SPC growth with self-renewal

    The association of proBNPage with manifestations of age-related cardiovascular, physical, and psychological impairment in community-dwelling older adults

    NT-proB-type natriuretic peptide (NT-proBNP) serum concentration can be transformed by simple formulas into proBNPage, a surrogate of biological age strongly associated with chronological age, all-cause mortality, and disease count. This cross-sectional study aimed to assess whether proBNPage is also associated with other manifestations of the aging process in comparison with other variables. The study included 1117 noninstitutionalized older adults (73.1 +/- 5.6 years, 537 men). Baseline measurements of serum NT-proBNP, erythrocyte sedimentation rate, hemoglobin, lymphocytes, and creatinine, which have previously been shown to be highly associated with both age and all-cause mortality, were performed. These variables were compared between subjects with and without manifestations of cardiovascular impairment (myocardial infarction (MI), stroke, peripheral artery disease (PAD), arterial revascularizations (AR)), physical impairment (long step test duration (LSTD), walking problems, falls, deficit in one or more activities of daily living), and psychological impairment (poor self-rating of health (PSRH), anxiety/depression, Mini Mental State Examination (MMSE) score < 24). ProBNPage (years) was independently associated (OR, 95% CI) with MI (1.08, 1.07-1.10), stroke (1.02, 1.00-1.05), PAD (1.04, 1.01-1.06), AR (1.06, 1.04-1.08), LSTD (1.03, 1.02-1.04), walking problems (1.02, 1.01-1.03), and PSRH (1.02, 1.01-1.02). For 5 of these 7 associations, the relationship was stronger than that of chronological age. In addition, proBNPage was univariately associated with MMSE score < 24, anxiety/depression, and falls. None of the other variables provided comparable performances. Thus, in addition to the known associations with mortality and disease count, proBNPage is also associated with cardiovascular manifestations as well as noncardiovascular manifestations of the aging process

    Mechanisms of endothelial cell dysfunction in cystic fibrosis

    Although cystic fibrosis (CF) patients exhibit signs of endothelial perturbation, the functions of the cystic fibrosis conductance regulator (CFTR) in vascular endothelial cells (EC) are poorly defined. We sought to uncover biological activities of endothelial CFTR, relevant for vascular homeostasis and inflammation. We examined cells from human umbilical cords (HUVEC) and pulmonary artery isolated from non-cystic fibrosis (PAEC) and CF human lungs (CF-PAEC), under static conditions or physiological shear. CFTR activity, clearly detected in HUVEC and PAEC, was markedly reduced in CF-PAEC. CFTR blockade increased endothelial permeability to macromolecules and reduced trans‑endothelial electrical resistance (TEER). Consistent with this, CF-PAEC displayed lower TEER compared to PAEC. Under shear, CFTR blockade reduced VE-cadherin and p120 catenin membrane expression and triggered the formation of paxillin- and vinculin-enriched membrane blebs that evolved in shrinking of the cell body and disruption of cell-cell contacts. These changes were accompanied by enhanced release of microvesicles, which displayed reduced capability to stimulate proliferation in recipient EC. CFTR blockade also suppressed insulin-induced NO generation by EC, likely by inhibiting eNOS and AKT phosphorylation, whereas it enhanced IL-8 release. Remarkably, phosphodiesterase inhibitors in combination with a β2 adrenergic receptor agonist corrected functional and morphological changes triggered by CFTR dysfunction in EC. Our results uncover regulatory functions of CFTR in EC, suggesting a physiological role of CFTR in the maintenance EC homeostasis and its involvement in pathogenetic aspects of CF. Moreover, our findings open avenues for novel pharmacology to control endothelial dysfunction and its consequences in CF

    Ag nanoparticle-based inkjet printed planar transmission lines for RF and microwave applications: considerations on ink composition, nanoparticle size distribution and sintering time

    Sintering of Ag Nanoparticle (NP) - based inkjet printed tracks is a crucial process for the next-generation digitally printed electronics. In particular, while the digital printing, as additive technology, is now well settled for what concerns either DC or signal applications both on rigid and on flexible substrates, this technology has not been demonstrated yet in the RF or microwave field, and a few works appear considering vacuum-evaporated films, screen-printed pastes or inkjet printed inks. We studied the effects of both ink composition and thermal profile on the resulting electrical properties, performing real-time resistance acquisition (DC) and post-annealing microwave measurements. We tested ink compositions featuring both different NP size distributions and different phase compositions, including a pure solvent/salt/metal one and a solvent/salt/metal/polymer one, resulting in a peculiar mass distribution and heat diffusion. The composition strongly affects the onset of electrical percolation and the final resistivity; on the contrary, the heating rate can either have an effect on electrical properties or not depending on the composition.
